Purification Method:
This monoclonal antibody was purified using multi-step affinity chromatography methods such as Protein A or G depending on the species and isotype.
Antigen Distribution:
Mouse CD80 is expressed on Activated B-Cells, Activated T-Cells, macrophages, and dendritic cells.
Background:
CD80 (B7-1) is a type I integral membrane protein related very closely to CD86 (B7-2). A main function of CD80 is the costimulation of T-cells through ligation of CD28 and CD152 (CTLA-4). T-cells activate with binding of CD28 to CD80 and binding of CD80 to CTLA-4 results in inhibition of T-cells. The clone 16-10A1 has the ability to block the costimulatory activity of CD80 while exhibiting no effect on the activity of CD86.

Application Notes:
Flow Cytometry: It is recommended to use the indirect method for signal enhancement when enumerating cells expressing CD80. A suggested method would be to stain cells expressing CD80 with <0.5 µg per 100 µl of whole blood (or per 1.0 x 10^6 cells in a 100 µl total staining volume). Each investigator should determine their own optimal working dilution for specific applications.
